Phosphatase Classification: Fold CC1

Fold CC1 has a single superfamily CC1.

Structure

We carried out structural alignment analysis of the crystal structures of five phosphatases of different families (Paladin and OCA has no structure), including human DUSP3 (DSP), PTEN (PTEN), PTPN1 (PTP), MTMR2 (myotubularin) and yeast Sac1p (Sac) (family names in parentheses). We compared the secondary structures and identified the common and family-specific secondary structures (SSs).

All the five phosphatases share the same SSs combination of BBHBBHBHHH (B - sheet, H - helix). The phosphatases have different N-terminal SSs. All phosphatases ends at the same SS, helix 5 (H5), except MTMR2, which has additional 4 helices at C terminus. The sheets and helices are inserted at:

  • B2-H1: Sac1p
  • H1-B3: MTMR2
  • B3-B4: PTPN1, MTMR2, Sac1p
  • B4-H2: PTPN1, MTMR2
  • H2-B5: Sac1p
  • H3-H4: MTMR2

We are uncertain about whether Sac1p has helix 5, because the region is not included in the available crystal structures (3LWT and 4TU3). However, we know the region is necessary for its catalytic activity (see Sac).
