PTPN14 subfamily technical notes
PTPN14 is generally poorly conserved between the FERM and phosphatase domains, but does have a number of motifs that are highly conserved between vertebrate members (PTPN14 and PTPN21) and invertebrate members. From N- to C-termini, these motifs are:
- P[st]YRp[ats]PDY[de].~
- SQPE~
- RPPY....sSTPdLa
- S[sn]PDLVS
Letters in square brackets are optional, lowercase is less conserved ~=small hydrophobic. Note the similarity between motifs 3 and 4.
Motif 3 includes a PPxY motif that interacts with WW domains, and is known to be involved in bindings the WW domains of KIBRA (WWC1) and YAP. A second PPxY motif is found in vertebrate and some invertebrate forms, with a consensus of PPxYPG.
